Alternate gram staining technique using a fluorescent lectin.

Fluorescence-labeled wheat germ agglutinin binds specifically to N-acetylglucosamine in the outer peptidoglycan layer of gram-positive bacteria. The peptidoglycan layer of gram-negative bacteria is covered by a membrane and is not labeled by the lectin. By exploiting this phenomenon, an alternative Gram staining technique has been developed.

A new technique for Gram staining paraffin-embedded tissue.

Five techniques for Gram staining bacteria in paraffin sections were compared on serial sections of pulmonary tissues from eight bacteriological necropsies. Brown and Hopp's method was the most satisfactory for distinguishing Gram-positive and Gram-negative bacteria. However, this method cannot be recommended as the preparations were frequently overstained, and the Gram-negative bacteria were s...

Ziehl-Neelsen Staining Technique Can Diagnose Paragonimiasis

BACKGROUND We evaluated the Ziehl-Neelsen staining (ZNS) technique for the diagnosis of paragonimiasis in Laos and compared different modifications of the ZNS techniques. METHODOLOGY WE APPLIED THE FOLLOWING APPROACH: We (1) examined a paragonimiasis index case's sputum with wet film direct examination (WF) and ZNS; (2) re-examined stored ZNS slides from two provinces; (3) compared prospectiv...

Can a Century Old Experiment Reveal Hidden Properties of Water?

In 1893 Sir William Armstrong placed a cotton thread between two wine glasses filled with chemically pure water. After applying a high voltage, a watery connection formed, and after some time, the cotton thread was pulled into one of the glasses, leaving a rope of water suspended between the two glasses. Although being a very simple experiment, it is of special interest since it comprises a num...

Golgi Staining Technique

The Golgi staining technique, also called the black reaction after the stain's color, was developed in the 1870s and 1880s in Italy to make brain cells (neurons) visible under the microscope [3]. Camillo Golgi developed the technique while working with nervous tissue, which required Golgi to examine cell structure under the microscope [3]. Golgi improved upon existing methods of staining, enabl...
